The amount of antibody used is mainly related to the volume of fluid and the number of cells. If the amount of cells is small, the whole system can be halved, that is, 50
μl
cell suspension + 2.5
μl
flow-through antibody. However, if there are very few target cells in the sample, a reduction in the amount of cells may have an effect on the results, resulting in only a small signal being detected.
