basic program Materials 小鼠腹腔巨噬细胞（单 元 6.1) D M E M - 5 : 含 5 % (W V ) 热灭活 FBS (HyClone) 的 D M E M (GIBC0 / B R L ) lmmol/L iVG 单 甲 基 乙 酸 精 氨 酸 （N G-monomethyl-L-arginine acetate， L-N M A ; Calbiochem 或 Research Biochemicals)， PBS 配制， 保存 小鼠重组7-干 扰 素 （IFN-7, Genzyme) 0111: B 4 型大肠杆菌LPS (Sigma) ，肿瘤坏死因子a (T N F -cx; Genzyme) 或其他 激活剂 2mmol/L N a N O 2 (Sigma)， D M E M -5 配制， 4〇 C 保存 V Griess 试剂 12m m X 75m m 带盖聚丙烯塑料细胞培养管（Falcon) 9 6 孔平底细胞培养板（Costar)
1 . 收 集 小 鼠 腹 腔 巨 噬 细 胞 （单 元 6.1)，用 D M E M - 5 调 整 细 胞 浓 度 至 I X IO6个细 胞/m l 。操作非 贴 壁 型 巨 嗤 细 胞 时 ：在 12m m X 75m m 聚 丙 烯 细 胞 培 养 管 中 加 入 0.5m l 细胞悬液。操作贴壁型巨噬细胞时：在 9 6 孔平底培养板中，加 入 0.1m l 细胞 悬液。如下设置实验组和对照组，每 组 2〜3 复孔： 第 1 组 ：未刺激组 第 2 组 ： IFN-7刺激组 第 3 组 ： LPS (或其他激活剂，如 TNF) 刺激组 第 4 组 ： IFN-7 和 LPS联合刺激组 第 5 组 ： 50〜25〇 iumol/L L-N M A 、 IFN-7 和 LPS 联合刺激组 2 . 操作贴壁型巨噬细胞时：将 9 6 孔细胞培养板置37°C ， 5 % C 0 2细胞培养箱中培养1〜 2h ，使巨噬细胞贴壁。然 后 用 D M E M -5培养基轻轻洗涤细胞2 或 3 次。 3 . 在 第 5 组中加入L -N M A ，终浓度为50〜25(V m o l /L 。 4 . 在 第 2、 4 、 5 组中分别加入IFN-7 (使巨噬细胞致敏），终浓度均为0.1〜l 〇 U/ml。 5 . 在 第 3、 4 、 5 组中分别加入LPS (使巨噬细胞激活），终浓度均为2〜10ng/m l 。 6 . 细胞培养24〜72h 后 ，取各组上清SOiUl分 别 加 入 9 6 孔平底板中。 7 . 将 N a N O 2溶 液 系 列 稀 释 （终浓度从1〜125Mmol/L ，各 取 50M1加 入 9 6 孔平底板中）。 8•在待测孔和N a N O 2标准孔中加入50jul Griess试剂 。最 好 先 加 入 萘 乙 二 胺 （naphthylenediamine) ，然后再加入氨基苯磺酰胺（sulfanilamide)。 9 . 用酶联仪检测N a N 0 2# 准孔和各待测孔的光吸收值，测量波长为550n m 。用 N a N O 2 标准孔的光吸收值和对应的浓度做一标准曲线（在 〇 〜 范 围 内 应 呈 线 性 相关）。用此标准曲线计算出各待测样品的N 〇 2— 值 。结 果 以 N 0 2— 浓 度 值 （Mmol/L ) 或标准化换算后的N O 「 摩尔值表示，即 IO5个 细 胞 或 I m g 蛋 白 样 品 的 N O 「摩尔数 (nmol/105个细胞或nmol/m g 蛋白）。 本 方 法 检 测 N O 「灵敏 度 为 l; xmol/L左 右 。
